You can but unless you are ewiped to do agar work it is a struggle to get a clean culture. Rub the cap with alcohol. Full sanitary procedure should be followed. A syringe can be squirted in the gills and caught in a shot glass, but just pulling a lamallae and putting it on agar and an early transfer of the first growth to a new dish is the standard.
Ultra low-tek= syringe to the gills and dumped in a bag of straw/cowshit outdoors. If it freezes where you live it'll die every winter but you can do it again in the spring. Outside there's a sort of symbiosis where so many contaminants are competing for real estate that they battle each other. Inside that technique is Trichoderma city with Pseudomonas as a suburb
